David Sinclair has a Ph.D. and is a professor in the Department of Genetics and co-Director of the Paul F. Glenn Center for the Biology of Aging at Harvard Medical School. Sinclair is enthusiastic
about the science of NAD boosters, like NMN, and their promise of improving human health. Furthering that science and the prospect of cellular age reversal is his life’s work. As part of this endeavor, he also supports other researchers perform clinical trials to address medical conditions such as glaucoma, kidney failure, frailty, and rare diseases like Friedreich’s ataxia. Sinclair is best known for his work on understanding why we age and how to slow its effects which lead to a very fascinating conversation on the Joe Rogan Experience podcast episode #1349 (2h 19min). Below is the key points from the conversation, although it is highly recommended to listen to the episode in full.
Key Points
- Sirtuins is a set of genes that controls aging. There are 7 of them in our body. They protect the body from deterioration and disease.
- Resveratrol: found in red wine, is like an accelerator for the sirtuin genes.
- Nicotinamide mononucleotide (NMN): the fuel, without NMN the resveratrol will not work.
- Nicotinamide riboside (NR): is a supplement that raises the levels of nicotinamide adenine dinucleotide (NAD+). Sirtuins are a class of genes that need NAD+ to work. Sirtuins do not protect the body without high levels of NAD+.
- NMN & NR both boost the body’s levels of NAD+ up to youthful levels again. Clinical trials have shown that this will raise levels of NAD+.
- Nicotinamide adenine dinucleotide (NAD+): is thought not be well-absorbed in the body, thus it is better to raise NAD+ levels indirectly by taking NMN. NAD+ is critical for everything from energy and metabolism and even sleep, it also replenishes the blood vessel lining.
- Metformin: originally a prescribed drug for diabetes. However, in studies of 10 000 people or more, it has been shown that people (even if they do not have diabetes) are protected against cancer, heart disease, Alzheimer’s and diseases typically associated with aging.
Sinclair’s book from Lifespan: Why We Age―and Why We Don’t Have To (2019) is highly recommend for those who want to go a bit more in-depth.
David Sinclair’s personal regime:
- NMN or NR: 1 gram in the morning (along with yoghurt).
- Resveratrol: 500 mg in the morning.
- Metformin: need prescription. 1 gram before bed.
- Alternative: Berberine – no prescription needed.
